The elevated tracks are designed for a top speed of 110kph, but during the test the train kept to under 40kph.
It stopped at the station in front of the Saigon Hi-Tech Park for passengers to take a five-minute tour.
"The test run marked the transition from the stage of construction to the stage of testing and getting it ready for commercial operation," a spokesperson for the HCMC Management Authority for Urban Railways (MAUR) said.
The line has 17 Japanese-made trains that can carry 930 passengers.
Since August this year all 17 have been tested at the Long Binh depot in Thu Duc City.
Following 10 years of construction the first metro line, with three underground and 11 elevated stations, is around 93% complete.
It is expected to begin operations in the fourth quarter of next year.
MAUR said trains would next be tested on the entire 19.7-km route between Ben Thanh and Suoi Tien next year.
